The Dutch giants are holding out for at least £10m for the 23-year-old, who has stepped up for Ajax since Thomas Vermaelen left for Arsenal last summer.
Liverpool have also monitored the versatile Belgium international, who can also play in midfield, but Everton have made the first move as David Moyes looks to strengthen his defensive options.
The Toffees have struggled to adequately replace Joleon Lescott, who quit Goodison Park for Manchester City last summer, and Moyes has also tracked Torino's Angelo Ogbonna.
